Osteoarthritis susceptibility 1

Common Name(s)

Osteoarthritis susceptibility 1

Osteoarthritis (OA) is a degenerative disease of the joints characterized by degradation of the hyaline articular cartilage and remodeling of the subchondral bone with sclerosis ({18:Meulenbelt et al., 2006}). Clinical problems include pain and joint stiffness often leading to significant disability and joint replacement. Osteoarthritis exhibits a clear predilection for specific joints; it appears most commonly in the hip and knee joints and lumbar and cervical spine, as well as in the distal interphalangeal and the first carpometacarpal (base of thumb) and proximal interphalangeal joints of the hand; however, patients with osteoarthritis may have 1, a few, or all of these sites affected ({29:Stefansson et al., 2003}). According to a conservative estimate, greater than 70% of the population of the United States at age 65 years is affected by the disease, reflecting its age dependence. Genetic Heterogeneity of Susceptibility to Osteoarthritis Susceptibility to osteoarthritis has been associated with variation in other genes: OS2 ({140600}) with variation in the MATN3 gene ({602109}) on chromosome 2p24; OS3 ({607850}) with variation in the ASPN gene ({608135}) on chromosome 9q22; and OS5 ({612400}) with variation in the GDF5 gene ({601146}) on chromosome 20q11. Other susceptibility loci for osteoarthritis have been mapped to chromosomes 2q33 (OS4; {610839}) and 3p24 (OS6; {612401}).

Association between transforming growth factor-beta 1 gene single nucleotide polymorphisms and knee osteoarthritis susceptibility in a Chinese Han population.

Author(s): Ning Lu, Jianwei Lu, Chenhe Zhou, FuHua Zhong

Journal: J. Int. Med. Res.. 2017 Oct;45(5):1495-1504.

Objective To investigate associations between single polymorphisms (SNPs) rs1800469 and rs1982073 in the transforming growth factor-β1 gene ( TGF-β1) and knee osteoarthritis (OA) susceptibility in a Chinese Han population. Methods TGF-β1 rs1800469 and rs1982073 were genotyped in ...

Functional characterisation of the osteoarthritis susceptibility locus at chromosome 6q14.1 marked by the polymorphism rs9350591.

Author(s): Katherine Johnson, Louise N Reynard, John Loughlin

Journal:

The arcOGEN genome-wide association study reported the rs9350591 C/T single nucleotide polymorphism (SNP) as marking a region on chromosome 6q14.1 that is associated with hip osteoarthritis (OA) in Europeans, with an odds ratio (OR) of 1.18 and a p-value of 2.42 × 10(-9). rs9350591 ...

Association of a functional microsatellite within intron 1 of the BMP5 gene with susceptibility to osteoarthritis.

Author(s): James M Wilkins, Lorraine Southam, Zehra Mustafa, Kay Chapman, John Loughlin

Journal:

In a previous study carried out by our group, the genotyping of 36 microsatellite markers from within a narrow interval of chromosome 6p12.3-q13 generated evidence for linkage and for association to female hip osteoarthritis (OA), with the most compelling association found for a marker ...
